Effect of Number of Air exchanges , sucrose and BA on Shoot Proliferation and Growth of ¢¥ Campbell Early ¢¥ Grape (Vitis hybrid) Plantlets




Abstract
The effects of 6-benzylaminopurine (BA) with or without sucrose (30 g¡¤L^(-1)) and the number of air exchanges (1.8 and 0.8 h^(-1)) on shoot proliferation and growth of grape plantlets grown under photoautotrophic or photomixotrophic culture conditions were investigated. Multiple shoots were successfully induced in all media containing 1 §·¡¤L^(-1) BA. The number of shoot increased more than four times as compared with non-BA treatment when the number of air exchanges was increased under photomixotrophic condition. The numbers of nodes were less than 3.0 in the BA treatments than 5.3 in the photoautotrophic condition without BA. Total dry weights were below half in the BA treatments than in the increased number of air exchanges under photomixotrophic conditions without BA. Hyperhydric plantlets occurred remarkably in the treatment of photomixotrophic condition with the decreased number of air exchanges and BA. A net photosynthetic rate per plantlet under photoautotrophic and photomixotrophic conditions with the increased number of air exchanges without BA was about 5 times higher than that of the decreased number of air exchanges under photomixotrophic condition with BA. When cultured under BA-free photoautotrophic conditions with the increased number of air exchanges, stomata showed more elliptical compared with those in control, photomixotrophic control conditions with the decreased number of air exchanges and BA. Abnormal stomata such as large or spherical types were also observed in photomixotrophic conditions with BA with the decreased number of air exchanges. BA that was added in the medium was effective in inducing multiple shoots, but depressed shoot and root growth. On the other hand, the growth was promoted under photoautotrophic and photomixotrophic conditions with the increased number of air exchanges. These conditions could be favorable for micro-proliferation with node (in vitro cutting) of grape.
